In diabetic models, SOD mimetics may play a role in improving endothelial dysfunction. For example, tempol (Figure 10), improves insulin sensitivity and decreases oxidative stress in obese Zucker rats as a model of type 2 diabetes.79 Tempol also reduces oxidative stress and improves endothelial dysfunction in streptozotocin-induced type 1 diabetic rats.80 NADPH oxidase is the main source of superoxide production following AT-II stimulation and mice lacking the p47phox subunit of NADPH oxidase have an attenuated hypertensive response to chronic AT-II infusion.81 Mice lacking the gp91phox subunit also have an attenuated hemodynamic response to acute AT-II.82 Thus, inhibition of NADPH oxidase may be a novel approach to improve endothelial function and lower blood pressure in hypertension.
