DA-releasing agents, 5HT2 agonists, and NMDA receptor antagonists are all effective in a variety of rodent assays. Most notably, all three classes of compound disrupt PPI, and this disruption is responsive to atypical antipsychotics.47 Since a large body of evidence suggests a dysregulation of each of these neurotransmitter systems in schizophrenia, the pharmacological disruption of PPI provides an animal model with considerable construct validity. This combined with the face validity of the PPI paradigm and the suggested predictive validity provided by back-validation studies with atypical antipsychotics provide support for this as a relevant model of positive symptomatology. However, an important caveat exists in the fact that the disruptions are pharmacological in nature. It is not unexpected that a DA antagonist reverses the effects of a DA-releasing agent, and given the incomplete understanding of the pathophysiology of schizophrenia a purely pharmacological disruption of the model cannot be ruled out, even with a truly novel NCE. It is therefore important to interpret these studies with care and to rely on more than one pharmacological tool to induce disruption of behavior.
