There is no model of migraine. However, there are model of parts of the pathophysiology in animals and in humans.40 The essential issue is: what is one trying to do? If the question is to screen or evaluate new chemical entities (NCEs) for migraine, then basic animal studies are used. For exploration of identified questions in humans, there are now models of trigeminal function in humans that can allow, for example, dose selection for further clinical trials. A list of models is shown in Table 2. A case history example would be the development to proof-of-principle of the potential for adenosine A1 receptor agonists in the acute treatment of migraine (see Section 6.16.7.4).
