Neuregulin

Neuregulin (NRG1) on chr8p13 contains a core epidermal growth factor (EGF) domain and activates Erb-3 or -4 receptor tyrosine kinase.30 It is involved in migration of GABAergic interneurons, signaling between axons and Schwann cells and the interactions between NMDA receptors and PSD-95. NRG-1 knockout mice show symptoms of schizophrenia. While 17 of 21 genetic association studies showed polymorphisms in NRG-1 associated with schizophrenia, the majority lie in noncoding regions of the genome. The NRG-1 genotype has been associated with the therapeutic response to clozapine in a Finnish population. The evidence for the involvement of NRG-1 in schizophrenia is typified as 'substantial but not incontrovertible'30 and there have been studies with failure to replicate.31
