Thiazolidinediones are rapidly absorbed, with peak serum concentrations of rosiglitazone occurring within 1 h and pioglitazone within 2 h of oral administration. Administration with food does not prevent drug absorption, but may delay the time to peak serum concentration. Both rosiglitazone and pioglitazone are avidly protein-bound, primarily to serum albumin. The plasma half-life for rosiglitazone is 3-4 h and it is extensively metabolized. Pioglitazone has a halflife of 3-7 h and it is metabolized by hydroxylation and oxidation. The active, but less potent, metabolites have half-lives of 16-24 h.
