Determination of Storage Conditions

Rat liver was homogenised in 50 mM sodium phosphate buffer, pH 7.0, containing 20 mM KC1 by five passes of a Teflon pestle in a Potter-Elvehjem glass homogenizer at 500rpm. The crude homogenate was centrifuged for 60min at 20,000 x g at 4°C and the supernatant desalted on a Sephadex G25 column. Various compounds were added to the supernatant, which was stored at 4°C, -20 °C and -80 °C. After one and two weeks, y-BBH activity was determined and compared with the activity in the fresh supernatant.
