Polyethylene glycol PEG in wash medium

Autoclave 10 g PEG 1500 (Aldrich) in a Wheaton glass bottle and allow to cool. Before it solidifies, add 10 ml unsupplemented DMEM (wash medium) and distribute 2-ml aliquots into 4-ml sterile polypropylene tubes. Cap tightly and store at room temperature.

Because PEG precipitates proteins, wash medium should not contain FBS. Not all batches of PEG give identical results; thus, screening commercial lots may be required. Good fusion results have been obtained with batches kept as long as 4 weeks; prolonged storage has not been tested.
